    “Research Breakdown on Tongkat Ali - Examine”

    In individuals with late-onset hypogonadism, Longfolia (dosed as a 200mg water soluble extract) is effective in enhancing libido and serum testosterone levels.[[44]](https://examine.com/supplements/tongkat-ali/research/#ref-44) This study ran 4 years at hypogonadism clinic in Malaysia and evaluated 320 men with a serum testosterone below 6umol/L (clinical hypogonadism) and noted that over a period of 1 month the average testosterone levels increased from 5.66+/-1.51umol/L to 8.31+/-2.47umol/L, an average 46% increase; no placebo group was used in this study.[[44]](https://examine.com/supplements/tongkat-ali/research/#ref-44) A few studies[[45]](https://examine.com/supplements/tongkat-ali/research/#ref-45)[[46]](https://examine.com/supplements/tongkat-ali/research/#ref-46) cite up to 4 other articles presented by the author M.I Tambi at various conferences that cannot be accessed online. M.I Tambi is not the patent holder of the extract used (LJ100)[[12]](https://examine.com/supplements/tongkat-ali/research/#ref-12) and a later independent trial in otherwise healthy men failed to find an increase in testosterone.[[28]](https://examine.com/supplements/tongkat-ali/research/#ref-28) The evidence in humans that are also infertile (hypogonadic) supports a small increase in testosterone, but the lone study in otherwise healthy humans without problems has failed to find an increase in testosterone 7.2 Estrogen Two quassinoids from Eurycoma, 13α,21-Dihydroeurycomanone and Eurycomanone have been tested in a 17α-ethynylestradiol induced uterotrophic assay (an in vivo test for antiestrogenicity[[47]](https://examine.com/supplements/tongkat-ali/research/#ref-47)).[[48]](https://examine.com/supplements/tongkat-ali/research/#ref-48) 2.44umol/kg was able to induce anti-estrogenic effects when injected, and although both showed effects Eurycomanone was more potent and had a similar efficacy to Tamoxifen at the same molar dose. However, due to low oral bioavailability and poor eurycomanone content in Eurycoma, these effects may not be seen in vivo.[[14]](https://examine.com/supplements/tongkat-ali/research/#ref-14) Based on a molar mass of 408.39916,[[49]](https://examine.com/supplements/tongkat-ali/research/#ref-49) this equates to a 996mcg injection of Eurycomanone (equivalent to 906mcg Tamoxifen), and its bioavailability of 10.5%[[14]](https://examine.com/supplements/tongkat-ali/research/#ref-14) would mean about ten-fold the dose required to be practically bioequivalent; and 30mg Tamoxifen would theoretically be bioequivalent to about 313mg Eurycomanone (close to 20g of the plant's dry weight). These effects may underlie some anti-cancer effects of Eurycoma, as it has been found to inhibit proliferation of MCF-7 breast cancer cells in vitro, which are highly estrogen responsive.[[50]](https://examine.com/supplements/tongkat-ali/research/#ref-50) Additionally, these anti-estrogenic effects may be a mechanisms for increasing spermatogenesis (production of sperm) in males as one study in estrogen-treated male rates indicated that a low dose of 8mg/kg bodyweight Eurycoma Longifolia given for 14 days was able to abolish the suppressive effects of estrogen treatment on spermatogeic cell count and sperm count (by normalizing them to control levels), and that Eurycoma in control without estrogen further increased sperm count; no abverse effect of estrogen was noted on sperm motility although Eurycoma increased this parameter.[[33]](https://examine.com/supplements/tongkat-ali/research/#ref-33)
